Preservation of primate ES/iPS cells and vitrification freezing methods

 
 
 

Comparison

Human iPS cells were cryopreserved in existing vitrified freezing solution (DAP213) and this product, and the colony formation rate on the day after thawing and the number of cells after 4 days (recovery rate) were compared with the non-frozen group. The system frozen with this product clearly showed a higher protective effect.

Thawed iPS cells were cultured on untreated plates to produce embryoid bodies, and then cultured on normal culture plates for one week. Proteins derived from trimeric embryoid cells were detected.
 
 

Outline operation

※Perform preliminary testing in advance in the target cell.
 
【Freezing method】
  1. Prepare liquid nitrogen in a clean bench.
  1. Primate ES / iPS cells are detached in colony condition in a detachment solution (0.25% trypsin/1mg/ml collagenase IV/PBS solution).
      • At this time, a 60 mm dish can be used to freeze confluent cells to 1-5 vials.
  1. Centrifuge the cells collected in step 2 and remove the medium. If multiple cells are to be frozen here, keep them ice-cooled and perform the following operations one at a time.
  1. Add 200 μl of the product, pipet well, cover the vial, and immerse the entire vial in liquid nitrogen as soon as possible (within 1 minute).
      • If the liquid remains clear, vitrification has been successful.
  1. Store in a liquid nitrogen tank or - 130°C deep freezer.
 
【Thawing methods】
  1. Prepare medium warmed at 37°C and prepare the required number of centrifuge tubes containing 9 ml of medium.
      • Thawing should be done one at a time.
  1. Vials of cells frozen in this product are transported to a clean bench in liquid nitrogen in a Dewar bottle or similar container.
  1. Open the lid of the removed vial and add 1 ml of warmed medium quickly and dissolve by pipetting.
  1. Transfer the entire volume of 3 into a centrifuge tube containing 9 ml of medium prepared in 1, centrifuge and wash.
  1. Seed and culture on feeder cells.
